- Broadly Neutralizing Antibodies Are Able to Recognize Multiple Strains of HIV at Once
Antiretroviral therapy has had an enormous impact on treating HIV infections around the world. The millions of people currently taking these treatments under medical supervision can reasonably expect to reduce their viral loads to undetectable levels, eliminate the risk of transmission and live a normal life span. However, antiretroviral therapy is not without shortcomings. People need to take these medications regularly for life, and low compliance can lead to drug resistance. There is a promising new option on the horizon. I am a researcher who studies AIDS treatments, and I believe that monoclonal antibodies could become game-changers for the treatment of HIV infections. HIV poses a challenge to the immune system. HIV presents challenges to antibodies Antibodies are proteins that serve as major players in the immune system’s response to pathogens, which cause disease, and allergens, which cause allergic reactions. Antibodies recognize specific markers, or antigens, on a potentially harmful substance and help the body eliminate it. Over the past few decades, researchers have been able to isolate individual antibodies specific to the individual pathogen or allergen they are meant to attack. With this advance, monoclonal antibodies made in the lab have become a major segment of the pharmaceutical industry. You can see numerous ads on TV or in magazines promoting monoclonal antibodies to treat osteoporosis, autoimmune disorders and various types of cancers. Antibodies can also be used to treat viral infections, including COVID-19. But using antibodies gets more complicated with HIV, the virus that causes AIDS in people. One reason is that HIV has an enormous number of variants circulating across the world and even within a single infected individual. In fact, the genetic variation of HIV within a single patient exceeds the genetic variation of all circulating influenza strains worldwide during an entire flu season. The immune system of an individual infected with HIV creates antibodies to neutralize the virus. However, because these antibodies can usually recognize only one particular strain, they are unable to neutralize other HIV strains circulating in the population. Furthermore, HIV can mutate within an infected individual and escape antibodies specific to the variant causing the original infection. This ability to mutate and escape ongoing immune responses is a critical factor in the virus’s ability to continuously replicate, a hallmark of AIDS. It also makes it difficult to design an antibody treatment that can account for HIV’s enormous genetic variability. Monoclonal antibodies are used to treat many types of cancer. Broadly neutralizing antibodies show promise The discovery of rare individuals who make anti-HIV antibodies that can be effective against up to 80% of circulating strains, however, has boosted prospects for antibody treatments for HIV. These broadly neutralizing antibodies, or bnAbs, have seen impressive results. Monkey studies have found that a single administration of bnAbs can prevent infection from SHIV, the nonhuman primate version of HIV. One study found that two broadly neutralizing antibodies were able to reduce viral loads to undetectable levels in infected monkeys. In people, one study administering two bnAbs also saw suppression of HIV replication and nearly undetectable viral loads. One early-phase clinical trial in 2021 showed that one bnAb could potentially offer protection against HIV infection. Long-term production of antibodies All the monkey and human studies mentioned above required re-administering the broadly neutralizing antibodies every three weeks or so to maintain effective concentrations. This runs into the same problem antiretroviral therapies face in terms of requiring the individual to retake the drug frequently for life. But researchers have found a potential solution. Using a small virus that doesn’t cause disease, called an adeno-associated virus, to deliver broadly neutralizing antibodies into the body can stimulate muscle cells to continually produce these antibodies. Because muscle cells have a prolonged life span and can last on average 10 to 16 years, they can be turned into factories that produce the antibodies essentially for life. Broadly neutralizing antibodies can target many HIV strains circulating around the world. One study my colleagues and I conducted using adeno-associated virus found that one monkey was able to produce these antibodies for over six years after a single injection. Another monkey that researchers dubbed the “The Miami Monkey” is considered functionally cured, meaning its viral loads have been at undetectable levels for prolonged periods even without continuous antiviral drug therapy. Two other monkeys have also been cured of their AIDS virus infections with this approach. Adeno-associated virus vectors for HIV antibody therapies still face one more hurdle: anti-drug antibodies, or antibodies the body produces in response to the antibodies in the treatment. Anti-drug antibodies can result when the body registers an antibody treatment as foreign and mounts an immune response against it, negating the treatment. They have also have caused problems for antibody treatments in cancer and autoimmune disorders. That may especially be the case for broadly neutralizing antibodies, which have unusual structures that deviate from what the body normally expects an antibody to look like. Researchers are working hard to develop simple and accessible approaches to help patients build tolerance to broadly neutralizing antibodies. Some of these approaches include delivering treatments to other areas that have greater immune tolerance than the muscle, such as to the liver and through the mouth. Words By Ronald C. - The Marker Key West Harbor Resort Honors National Key Lime Pie Day with New Turndown Service
On July 1, 2006 both the Florida House of Representatives and the Florida Senate passed legislation selecting the “Key lime pie” as the official pie of the state of Florida. Today, on National Key Lime Pie Day, The Marker Key West Harbor Resort launches a new Key Lime turndown service as the perfect bedtime amenity for guests. As guests return from their day exploring the beautiful Key West Seaport, an array of Key Lime amenities will be arranged at their bedside. Upon entering, a slice of not too sweet, not too tart, key lime pie with the perfect crust to filling ratio will serve as a perfect late night snack accompanied by a bottle of Sauvignon Blanc. This wine choice offers tasting notes of grapes that pair beautifully with the citrus aroma of Florida Key Limes and a dash of coconut. The service also includes Key Lime scented bath and shower amenities by Key West Aloe offering a zesty tropical escape. Key Lime-flavored mints will also be provided. This new turn down service is available upon request and will run through the rest of the year. Anyone who has had the pleasure of visiting Key West knows the significance Key Lime Pie has on the old town. It is highly likely that the first Key Lime Pie was created by "Aunt Sally," the cook of a ship salvager and Key West's first millionaire. Many of the men aboard his ships would have had access to the basic and non-perishable ingredients required for the early recipe: canned milk, limes and eggs. Since then, Key Lime Pie has become one of the most beloved dishes in The Sunshine State. By ML Staff. Images courtesy of The Marker Key West Harbor Resort.

- SLS Brickell's Fi'lia Relaunches with New Interior Design and Menu
Fi’lia, the beloved Italian-American restaurant at SLS Brickell, has relaunched with new lunch and dinner menus, fresh seasonal cocktails, a new curated wine list, daily Aperitivo happy hour, and stunning new designs all centered around Fi’lia’s wood-fired cucina and warm, family atmosphere. Concepted by Executive Chef Victor Rosado alongside gastronomy and mixology pioneers Carte Blanched, Fi’lia’s new culinary offerings draw inspiration from Italy, Miami and the Mediterranean, fusing traditional dishes with exciting new creations made with fresh, locally sourced ingredients and house-made doughs. Elegant primi piatti include ultra-tender octopus that is hand-massaged, broiled, and grilled with smoked pimento cheese, a flavorful beef carpaccio – available as an appetizer or a pizza – and grilled Italian sausage sourced from Miami’s own Proper Sausage, served with house-made ciabatta bread and zesty aioli. From the new “Chef’s Corner,” the colorful corn raviolo is a perfect medley of Florida-meets-Amalfi Coast flavors with fresh lobster and a decadent lobster sauce, while the truffle and mushroom risotto speaks to Chef Rosado’s love of seasonal truffles, which can be added to any dish upon request. Fi’lia’s new menus also feature a locally loved selection of grilled meats, vegetables, and seafood dishes designed to share, including whole branzino, the ever-popular roasted half-chicken, and rotating specials such as an epic Bistecca alla Fiorentina and a full rack of lamb. “While Fi’lia was already considered one of the best eateries in Brickell and well loved by a tremendous amount of Miami locals,” said Chef Rosado, “we’ve now taken it to an even higher level with our revitalized menus and sharp new interiors. It’s the kind of place that diners want to come back to again and again.” Chef Rosado, whose impressive resume includes stints at Jean-George’s Fern Restaurant and José Andrés’ Mi Casa and Bazaar Mar restaurants, often welcomes arriving diners from the open kitchen or makes a stop at each table to converse with guests. This sense of community is at the heart of Fi’lia’s relaunch and reflected in the restaurant’s warm, welcoming service, as well as its revamped dining room. Alongside the new menu items, Fi’lia unveils stunning white-meets-wood interiors framed by cozy banquettes, floor-to-ceiling windows, soft candlelight, and lush greenery — an idyllic setting for delicious, unhurried meals shared with family and friends, old and new. Outside on Fi’lia’s al fresco terrace, large groups can gather for birthdays, anniversaries, and work celebrations under the stars, right in the heart of Brickell. And for extra special occasions, an exclusive chef’s table offers a front-row seat to Fi’lia’s open kitchen. To start their gastronomic journeys, diners are invited to sip on newly designed craft cocktails, dreamed up by Fi’lia GM Cristian Munarriz and Chef Rosado. Unique libations like the Negroni Bianco and Truffle Old Fashioned nod to Fi’lia’s Italian roots, while Miami- and South American-inspired cocktails like Smoky Passion and Mojito del Sol incorporate tropical fruits like pineapple, passionfruit, and mango using an ancient Roman “shrub” technique for perfectly balanced cocktails that are sweet, tart, and totally refreshing. Alternatively, wine lovers will delight in Fi’lia’s brand-new wine list curated by the restaurant’s on-site sommelier, F&B Director Justin Wilson. With hand-selected varietals from the finest regions of Italy, as well as a handful of French and Californian wines, Fi’lia’s new menu encourages guests to expand their palates and discover trailblazing winemakers from around the world in a friendly, educational environment. - New Technology Brings Relief to People Suffering from Chronic Nasal Conditions
According to the American College of Allergy, Asthma & Immunology, more than 31 million people in the United States suffer from sinus infection (known as sinusitis) each year. These individuals experience frequent facial pain, pressure or congestion; discharge of mucus that is yellow or green; difficulty breathing through your nose; even teeth pain, loss of smell or taste senses, headaches, fatigue, sore throat or bad breath. By Andrew Rivera MD (pictured above, left) and Paul Foster (right), MD of South Florida ENT Physicians Andrew Rivera, MD and Paul Foster, MD of South Florida ENT have been in practice for years offering the entire spectrum of treatment options for these conditions. They have noticed a gradual trend toward more in-office treatments. “Plenty of people are simply unaware that their chronic conditions can be treated in the office depending on the severity of their disease,” said Dr. Rivera. “There’s no longer a need to suffer from annoying runny, stuffed up and irritated nasal passages. Our patients have enjoyed excellent results from these procedures.” According to Dr. Foster, most people try to solve their nasal and sinus problems by using over the counter remedies and rely on the help of treatments from urgent care centers and primary care physicians. “When these remedies are not sufficient, patients often do not know where to turn or how to alleviate their symptoms,” he said. “In the past, chronic nasal and sinus problems would potentially require invasive surgeries with general anesthesia and often significant downtime with lengthy recoveries. “Thanks to advances in technology, however, today we can offer our patients minimally-invasive interventions that provide relief from common chronic conditions with less risk and minimal down time.” Dr. Rivera stresses that common symptoms of sinusitis, such as an inflamed nose, sinuses and throat, lasting more than three months are cause for concern and medical intervention. “Anything that obstructs the air-filled pockets located behind the facial bones surrounding the nose can lead to a buildup of mucus, infection, and inflammation causing significant discomfort,” he said. “We offer several treatment options to our patients, depending on their individual conditions.” South Florida ENT offers the following procedures Balloon Sinuplasty When patient’s sinus passages are blocked and medicine alone has failed, sinus surgery, performed under general anesthesia. would have been traditionally required. More recently, a minimally invasive procedure, that can be done in the office, is designed to unblock and dilate the sinus openings. Balloon sinuplasty was created for patients suffering from chronic sinusitis, this is a minimally invasive procedure conducted with local anesthesia. Inflamed sinuses are unblocked in a way similar to the method used by cardiologists to open up blocked arteries using balloon angioplasty. Sinuses remain open after the procedure allowing for the return of normal drainage, and, most important, sinus pressure relief. “For patients who want to avoid general anesthesia and the downtime required for healing after traditional sinus surgery, office dilation is a great option,” said Dr.Foster. This three-step process involves inserting a balloon catheter into the inflamed sinus, then inflating the balloon to expand that opening. Any infection or fluid in the sinus is then suctioned out. The system is removed during the final step, leaving the sinuses open. This treatment method is recommended for patients who have failed to get relief from medications. This procedure involves the use of a navigation system that safely guides the instruments into the sinuses. It is used to visualize the exact location of the instruments in the patient’s nose with three-dimensional diagnostic imaging. “Minimally invasive sinus treatment with navigation has revolutionized how obstructed sinuses are treated,” added Dr. Rivera, “and has been able to improve the quality of life for people suffering from sinusitis.” Cryotherapy Often patients with runny noses find relief from nasal sprays, but for others such an approach is not enough. Other patients do not want to avoid the side effects of staying on medications for months or years. As knowledge has grown about the anatomy of the nose and the influence of the nervous system, new office treatments have been developed to treat chronic runny noses. Some of these procedures take as little as 30 seconds. Cryotherapy, for example, is a treatment targeted at freezing those nerves that send signals to the tissues in the nose to produce mucus. The freezing interrupts these signals and reduces the production of mucus significantly. This cool temperature treatment is a solution for people dealing with chronic rhinitis with symptoms such as runny nose, nasal congestion and postnasal drip. Patients who have failed to gain relief from nasal sprays, drops or medications are often candidates for this treatment. Local anesthesia is applied, and most patients can return to normal activity the same day. In addition to the several minutes used to numb the area, the treatment itself only takes 30 seconds for each side of the nose. Most patients notice improvement within two to six weeks with minimal downtime. Turbinate Coblation Patients with nasal blockage that has not responded well to nasal medications or allergy treatments may be candidates for the turbinate coblation. This procedure reduces the size of the inferior turbinate or the soft tissue of a structure in the nose. During the procedure anesthetic spray and decongestant spray are applied to each nostril. Cotton soaked in anesthetic and decongestant medication is placed in each nostril. Much like a dental procedure, small doses of anesthetic are applied to ensure the nasal area is numb. A coblation device connected to a radio frequency generator is placed into the nose. It has a small electrode at the end, which is inserted into the turbinate. Once in place, radiofrequency is applied approximately two to three times to the turbinate tissue through the electrode. “In our practice, we have seen countless patients improve their lives by ending months, even years, of irritating nasal conditions with these simple minimally invasive procedures,” said Dr. Foster. “Our procedures use highly sophisticated technology to minimize side effects and maximize results,” added Dr. Rivera. - L'Oréal Introduces Perso, a Home Personalized Beauty Device
This 6.5-inch tech device, developed by Guive Balooch's Tech Incubator, delivers personalized on-the-spot skincare and cosmetic formulas. By harnessing the power of Artificial Intelligence, the level of personalization will be optimized over time as the system gathers more data about our customer’s skin and personal preferences. “We are committed to making L’Oréal the leader in Beauty Tech – and Perso is the next step in that exciting journey”, said Nicolas Hieronimus, our Deputy Chief Executive Officer. New applications on the way At CES 2020, we unveiled Perso’s capabilities for skincare. However, the 3-in-1 device will also be able to create custom formulas for lipstick and foundation — which will be launched at a later date.“L’Oréal is one of the most trusted names in beauty, and with Perso, we are putting personalized technology directly into the hands of our consumers,” said Guive Balooch, Head of our Technology Incubator. “Perso uses AI to optimize the formulas and actually gets smarter as you use it.” How Perso works Perso creates personalized skincare formulas in four steps: 1 - Providing a personal skin analysis: Our customer will launch the Perso mobile app and take a photo with their smartphone camera. Thanks to the ModiFace technology we have acquired in 2018, their overall skin condition—including deep wrinkles, fine lines, the appearance of dark spots, and pore visibility – will be analyzed with the help of AI. 2 - Analyzing the Environment: Using Breezometer geo-location data, Perso takes into account the local environmental conditions that can influence the state of our customer’s skin. These might include weather, temperature, pollen, UV index, and humidity. 3 - Adapting to personal preferences: The next step is to enter personal skincare concerns into the Perso app, including fine lines, dark spots, pigmentation, pore size, radiance, and dullness. To customize the unique formula of moisturizer, serum, and under-eye cream even further, the preferred texture and hydration-level can also be added. 4 - Making a custom formulation and dispensing: All of this data helps to create a personalized blend of high-performance skincare, dispensed in a perfectly portioned, single dose at the top of the device for easy, clean application. The technology adjusts for morning and evening application, and the device features a detachable mirrored top so that our customers have the option of taking a single or larger dose with them on-the-go. Perso’s hardware features a patented motor system located at the top of the device which moves and compresses the formula from the cartridges at the base of the machine in an upward motion to the dispensing tray above. Looking ahead to new features Perso's future makeup offerings will be able to incorporate real-time trend information as well as color-matching technology. So, our customers would be able to design a lipstick shade to match their outfit or to opt for a color that is trending on social media at that moment. In the meantime, our focus will be on the launch of Perso in partnership with one of our leading skincare brands in 2021. For more info visit L'Oréal By ML Staff. Images courtesy of L'Oréal
- Renowned Swiss Beauty Brand, "timeblock," Launches in the U.S.
A transformative luxury beauty brand with a refreshing and revolutionary take on beauty, timeblock® has launched its award-winning solutions in the United States. The brand selectively sources natural ingredients from all over the world and uses Swiss-technology harvesting processes to ensure optimal herbal nutrient efficacy for its products, which include nutritional supplements and skincare products to combat the visible signs of aging. Beauty aficionados in the United States can now purchase the botanical anti-aging products on the timeblock® website, with nationwide shipping available. Also available for purchase in the U.S., the brand has a collection of aromatic diffusers. A winner of “Product of the Year” at the Global Health Asia Pacific Summit in 2019, the science-backed and evidence-based brand was developed over 20 years ago by a group of leading scientists in Switzerland, who set out to decrypt the secrets of Mother Nature. After experimenting for over seven years, they discovered that select plants and grasses grown at ultra-high altitudes and in precise environmental conditions and locations, such as the Himalayas, Brazil and Tibet, produce nutritional densities thousands of times higher than the same plants and seeds grown elsewhere. These nutrient-rich superfoods such as wheat grass, barley grass, shiitake mushrooms, tomatoes, grape seeds, green tea, marigold and algae, came to form the basis of the brand’s signature line, timeblock® Vital Aging Nutrition. Vital Aging Nutrition consists of daily daytime and nighttime supplements, Nutrition Day Complex and Nutrition Night Complex, that align with the philosophy that beauty starts from within. The Day Complex (green) is taken in the morning for energy and radiance throughout the day, while the Night Complex (red) is taken in the evening to promote rest and regeneration during the night. The effectiveness of the science-backed products was supported by the findings of the University of Vienna, who measured key aging markers such as DNA damage, inflammation, telomere length and cell reproduction in 120 participants taking timeblock® Vital Aging Nutrition supplements. At the end of the trial, results showed an astounding 23 percent reversal in biological age of participants within six months. Due to the overwhelming success of the Vital Aging Nutrition range and its scientifically proven results, the creators of timeblock® created a complete system that could target aging both inside and out. Thus, timeblock® Skin Care was created, with products including: 4 WEEKS Intensive Face Care A four-week intensive skin treatment consisting of a face care lotion that helps overnourished, tired and stressed skin help regain its natural regenerative ability. DAY Face Care A high-performance, super-antioxidant moisturizing lotion designed that uses apple stem cells to boost cell activity and protect the skin against environmental aggressors such as sun and pollution. NIGHT Skin Repair A serum with exceptionally high concentration polypeptides that supports skin renewal and regeneration during the body's natural rest and regenerative phase at night. Skincare products contain the anti-aging ingredient equol. More expensive than gold, equol is the most expensive active ingredient in the skincare market and a powerful skincare molecule that improves signs of aging. It also helps to accelerate cell renewal, protects against environmental aggressors, contributes towards a more even complexion and healthier skin, and moisturizes skin while simultaneously stimulating collagen and elastin production. Customers further looking to indulge in self-care can also purchase the Aromatic Ultrasonic Diffuser, a pineapple-shaped diffuser (available in gold, silver and rose gold) which uses ultrasonic technology to break down any mixture of essential oils and water to purify, humidify and refresh air. “Healthy aging innovation is the core of timeblock® as well as their commitment to creating a better quality of life and longevity for people globally,” said Dr. Alexander Hasleberger, Professor of Nutritional Science at the University of Vienna and former World Health Organization consultant. “Their products are elevating the boundaries of natural beauty through science and technology.” For more information on timeblock®’s products, as well as a breakdown of ingredients, visit www.timeblock.us.
